New mouse model suggests how Alzheimer’s causes dementia

Beta-amyloid plaque accumulation can damage the brain but is not sufficient to trigger the conversion of tau or the full neurodegenerative process. A second insult—the conversion of tau—driven by signaling events, may be required, implying that therapies targeting both amyloid and tau conversion, and doing so before tau changes occur, could be more effective.
